Comme cela avait été décidé, les musulmans partirent en direction de la vallée d’Uhud où campaient les Quraysh. Arrivé au mont Aynayn, qui supplante la vallée, il décida d’y cantonner certaines des troupes musulmanes. Celles-ci étaient quatre fois moins nombreuses mais leur confiance en la victoire était inébranlable. Combattants au nom de Dieu, les musulmans ne se préoccupaient guère de leur sort. Beaucoup se disaient que la meilleure chose qui pouvait leur arriver, c’était trouver la mort sur un champ de bataille en compagnie du Prophète (PSL). Lors de la réunion du conseil de guerre, Abdullah ibn Jubayr, presque en transe, s’était écrié : « Seigneur, je T’implore de me permettre de rencontrer nos ennemis demain, et de faire qu’ils me tuent, m’éventrent et me coupent le nez et l’oreille. Quand Tu me demanderas pourquoi ils m’ont fait cela, je Te répondrai : « Parce que je crois en Toi ! ». Les Quraysh étaient venus pour se venger de Badr, mais les musulmans, qu’ils attaquaient, étaient mille fois plus motivés.
Avant la bataille, Mouhamed (PSL) en vrai chef de guerre positionna ses troupes. Avec la supériorité numérique de leurs ennemis, il leur fallait être à la fois stratégiques et disciplinés. Ainsi, des archers furent placés sur un versant du mont Aynayn et chargés de protéger les musulmans à l’arrière. Autrement, ils devaient empêcher à l’ennemi d’encercler les musulmans. Ils ne devaient quitter cette position sous aucun prétexte. Les cavaliers des Quraysh, sous le commandement de Khalid ibn al-Walid, une fine lame au courage et à la perspicacité légendaires, étaient à l’affut.

Certains exégètes font état du corps de Hamza qui aurait été profané par Hind qui ne pardonnait même pas à la dépouille de celui qui avait tué et son père et son frère. 
Cette perte n’empêchait pas aux musulmans de dominer leurs adversaires et de leur marcher dessus. Surpris une nouvelle fois par cette détermination, les Quraysh commencèrent à battre en retrait. Nombre n’entre eux prirent la fuite. Le coup de Badr était sur le point d’être réédité. Cependant, le relâchement gagna les musulmans qui croyaient la victoire acquise. Aveuglés par un improbable triomphe qui se dessinait pourtant, ils se mirent à se préoccuper du butin, de ce qu’ils pouvaient prendre à l’ennemi. Cette déconcentration ne tarda pas à gagner les archers placés sur le mont Aynayn qui eurent le sentiment que la bataille était terminée. Ils descendirent sur le champ de bataille pour participer au ramassage du butin. Khalid ibn al-Walid ne se fit pas prier. Il engagea ses troupes dans la brèche et porta un violent assaut qui fit beaucoup de dégâts dans les rangs des musulmans. A lui seul, Khalid réussit à retourner la situation. Mouhamed (PSL) et ses troupes furent pris en tenaille. Les soldats Quraysh qui fuyaient reprirent courage et rebroussèrent chemin. La confusion était alors du côté des musulmans. Tout d’un coup, au paroxysme de la confusion, un cri résonna au-dessus de la mêlée : « Mouhamed (PSL) a été tué ! ». Aussi surpris que désarçonnés par cette annonce, certains se mirent à douter. D’autres partirent vers la montagne pour s’y refugier. Les Quraysh qui sentaient l’équilibre des forces tourner à leur avantage, prirent le parti de s’en prendre directement au prophète Mouhamed (PSL). Ils savaient qu’à ce stade de l’évolution de l’Islam, les musulmans ne pouvaient survivre à leur guide. Ils mobilisèrent ce qui restait de leurs forces à cette tâche dont l’accomplissement pouvait définitivement consacrer leur victoire et leur vengeance. Mais c’était sans compter sur certains musulmans qui malgré la déroute qui se dessinait, étaient prêts à se faire tuer à la place du Messager. Leur courage et leur détermination eurent raison des velléités offensives des Mecquois qui finirent par desserrer l’étau. Beaucoup de musulmans furent tués, de nombreux autres blessés. Le Prophète (PSL) lui-même reçut plusieurs coups.
«Et tout ce que vous avez subi, le jour où les deux troupes se rencontrèrent, c’est par permission d’Allah, et afin qu’Il distingue les croyants », (Sourate 3 – Verset 166).
